A Clinicopathological Study and Ki67Expression of Glioma in Jilin Province, North East of China
【摘要】：Background: After the establishment of relationship between angiogenesis and tumorgrowth by Folkman in1971, efforts are being made for treating cancer by targetingangiogenesis. Melanoma differentiation-associated-7(MDA-7) gene is a novel melanomadifferentiation associated gene that functions as a multi-modality anti-cancer agent.Manystudies have demonstrated it antiangiogenesis effect in different tumor models by variousroutes.
Objective: The aim of this study to demonstrate its antiangiogenesis effect throughintramuscular injection of MDA-7gene with AAV in lymphoma mouse model byimmunohistochemistry study measured by expression of cd31.
Methods: After successful establishment of lymphoma mouse model with singleintramuscular MDA-7gene therapy, after about7weeks cd31staining of lymphomatumor is examined for counting of microvessels density with control used as GFP vectors.
Results: Stastical analysis of microvessel density staining demonstratedsignificantly(p=0.027) diminution in tumors from mice administrated AAV8-MDA7/IL-24,which showed an average microvessel count of only11.42+/-2.8than the control goupAAV-GFP with the value of15.25+/-4.8.
Significance: This anti angiogenesis property of intramuscular AAV/MDA-7gene therapy in lymphoma mouse model provides a rationale for treatment of lymphoma infuture.
